GLP-1 drugs cut weight in adults without diabetes
At Jewish General Hospital and McGill University, researchers followed the evidence from 25,816 adults without diabetes—not through one new treatment, but across 38 randomized controlled trials. Their updated review found that GLP-1 receptor agonists and related coagonists produced significantly more weight loss than placebo.
The review was designed to capture the expanding range of these therapies, including newer agents and more evidence than the researchers' previous assessment. Among treatments currently available, tirzepatide produced the greatest weight loss. Some emerging experimental drugs were associated with reductions exceeding 20% of body weight.
The trade-off was familiar across the studies: gastrointestinal side effects were the most common adverse effects reported. At the same time, serious adverse events and deaths were rare, and the authors found no new safety concerns in the evidence they examined.
So what changes for people without diabetes and living with overweight or obesity? The findings point to a growing set of weight-loss options based on GLP-1 biology, with both injectable and oral treatments becoming available. The strongest results in the review belong partly to drugs that remain experimental, so they should not be read as established treatment outcomes.
The review measures effects across controlled trials rather than guaranteeing the same result for every patient. Its authors conclude that GLP-1-based therapies offer effective weight-loss options without diabetes, while the frequency of gastrointestinal effects remains a practical limitation.
