STMP1 helps protect aging hearts in laboratory models

Inside the heart’s energy factories, researchers from the Yong Loo Lin School of Medicine at the National University of Singapore found a small protein with a large job. When laboratory models lost STMP1, a protein only 47 amino acids long, their heart muscle cells developed mitochondrial damage, inflammation and weaker pumping.

The team used three-dimensional electron microscopy to examine the mitochondria. STMP1 helped maintain their cristae — folded inner membranes that support energy production. Without it, mitochondrial DNA leaked into the surrounding cell. The cell mistook that misplaced DNA for a sign of infection and activated cGAS–STING, an immune alarm that sustained inflammation and contributed to scarring and heart muscle cell death.

Signs of inflammation appeared before obvious heart failure. STMP1 levels fell specifically in cardiomyocytes, the cells that make the heart contract, and were also lower in human hearts affected by dilated cardiomyopathy. That links the laboratory mechanism to tissue from failing hearts, although it does not yet show that low STMP1 causes heart failure in patients.

The researchers tested two ways to interrupt the process. Gene delivery restored STMP1 in the heart, improving mitochondrial structure, reducing inflammation and restoring function in laboratory models. A drug that blocked STING also preserved heart function and reduced inflammation, cardiac stress and scarring — but only when given early, before substantial mitochondrial damage had developed.

The practical implication is a possible route to treating an underlying process rather than only the symptoms of age-related heart decline: restore STMP1, block the inflammatory pathway, or measure STMP1 to identify risk earlier. For now, the evidence remains preclinical. The approaches have not been tested in patients, and the NUS team plans larger, more disease-relevant studies before that step.
