Urolithin A boosts cyclosporine in rodent lupus study
At the University of Alabama’s Drug Research and Engineering for Advanced Medicine Laboratory, Dr. Ravi Kumar’s team tested an existing lupus drug beside a compound associated with food. In a female-rodent model of lupus nephritis, cyclosporine combined with synthetic urolithin A performed significantly better than either treatment alone.
The dietary connection is real but less simple than a spoonful of pomegranate. Gut bacteria can metabolize compounds from pomegranates, berries and nuts into urolithin A, yet only 30% to 40% of people naturally carry the specific microbiome needed to make it. The researchers therefore administered a synthetic version, giving the experiment a consistent dose.
Cyclosporine alone showed some benefit, consistent with its existing use in lupus treatment. Urolithin A alone had limited effect on disease progression. The approach was tested only in a preclinical rodent model, with females selected to reflect lupus’s higher prevalence in women than men.
So what changes, concretely? The result points to a possible way of supporting an established immunosuppressant with a compound already available in dietary supplements, rather than treating the supplement as a standalone lupus therapy. For patients, however, nothing changes in the clinic yet: prescribed medication should not be replaced, and adding a supplement requires medical advice.
Urolithin A has an FDA “generally recognized as safe” designation for use in commercial dietary supplements and functional foods, but that is not evidence of clinical benefit for lupus. Kumar’s stated aim is to replace anecdotal supplement claims with systematic testing. The combination now offers a practical candidate for further translational work, while human studies and clinical evidence are still needed.
