In hypertensive mice, drug makes the aorta 37% more flexible

For 28 days, researchers watched the blood pressure, heart rate and activity of 45 male mice. At the end of the study, ultrasound and tissue examinations found that the experimental compound Cmpd17b had lowered pressure and reduced damage in the kidneys, aorta and heart of hypertensive animals.

The drug targets formylpeptide receptors, or FPRs, switches found on immune, heart and kidney cells that help direct the inflammatory response. In mice with high blood pressure, Cmpd17b lowered mean arterial pressure by 6 mmHg. The effect built gradually, with no sudden fall after dosing, and was strongest during the mice’s dark, active phase. Mice with normal blood pressure saw no change.

The clearest result was in the large artery leaving the heart. Cmpd17b made the aorta 37% more flexible and reduced its wall thickness by 22%. It also cut fibrosis—the buildup of stiff scar tissue caused by chronic damage—in the kidneys, the aorta and the left ventricle, the heart’s main pumping chamber.

That matters because hypertension is not only a reading on a monitor. Nearly 1 in 8 people worldwide are affected, and long-term pressure can damage the heart, kidneys and blood vessels. The study tests a different strategy from broad anti-inflammatory drugs: rather than suppressing the entire immune system, it acts on FPRs involved in the healing phase of inflammation.

So what changes in practice? Nothing for patients yet: this was an early study in mice, and larger animal models plus human trials are still required. Cmpd17b also failed to improve heart pumping performance or fully restore smaller blood vessels. But the combination of a modest pressure reduction and measurable improvements in arterial flexibility and kidney scarring gives researchers a concrete lead for treating damage already caused by hypertension.
