A protein that normally sits at the heart of a cell's RNA-interference machinery has been turned into a tool that edits RNA. The result appears in a study published on 20 October 2026 in Nature Biotechnology, titled "Engineered human Argonaute 2 for targeted C-to-U RNA base editing". The protein is Argonaute 2 (AGO2), the catalytic engine of the RNA-interference pathway, the cell's system for regulating RNA. What the team shows is that this human enzyme can be redirected to change a single letter of RNA.

The method rests on guide RNAs. In the earlier preprint on bioRxiv, the team describes using guide RNAs that remodel the structure of the target RNA, which allows selective editing and reduces the enzyme's nuclease activity, meaning its tendency to cut the molecule. The system is called ALTER. The preprint reports that ALTER can act on a variety of targets. The chemistry it performs, a cytosine (C) turned into a uracil (U), is the change the Nature Biotechnology title names.

The central comparison comes from transcriptome-wide RNA sequencing, a method that reads the RNA present across a cell. There, the authors report lower levels of off-target editing than with tools built on Cas proteins, the enzymes behind CRISPR gene editing. Off-target means edits at sites other than the one intended. The team gives no percentage for the difference, and the excerpts available do not report one.

The approach is not the only one in its field. A separate paper, indexed in PubMed Central, converts the human enzyme ADAR2 into an enzyme that performs the same C-to-U change, and its title refers to efficient editing in vivo, that is, inside a living organism. The two projects take different human proteins as their starting point.

For laboratories that study RNA, the gain is a tool built from a protein the human body already makes, which the authors stress despite its role in RNAi. Research groups are the first to benefit, if the approach holds up in their hands. Neither publication gives a date for use beyond the laboratory, and the work remains at the lab stage.
