Still early: one eye injection plus camera goggles, for a disease that Nature Medicine says has no other treatment.

Ten people blinded by advanced retinitis pigmentosa each received a single injection into the eye. The injection carried a gene that tells cells to make Chrimson, a light-sensitive protein. Afterwards, patients wore goggles with a built-in camera that sends visual information to a portable processor. The study, published in the New England Journal of Medicine (NEJM) and led by Pitt scientists at the University of Pittsburgh School of Medicine with international collaborators, was built to answer one question first: is the treatment safe?

Retinitis pigmentosa can result from variants in over 100 genes, which is why gene-therapy approaches have had limited use. Nature Medicine notes that, in this condition, no other treatments exist. The team did not try to correct a faulty gene. The vector aims at ganglion cells in the retina and adds a light-sensitive protein, so the cells can respond to light, while the goggles supply the visual input.

Safety was the primary outcome, and the treatment was considered safe in all participants. The team tested three dose-escalation groups, at 5.0 x 10^10, 1.5 x 10^11 and 5.0 x 10^11 vector genomes per eye. Six patients showed a clinically meaningful increase in light sensitivity, and several improved at tasks such as detecting or locating objects. Visual signals were also detected in the brain. The findings extend an earlier single-patient result.

The numbers are small, and the study is open-label, meaning participants and clinicians knew which treatment was given. Nature Medicine describes the efficacy signals as early and points to opportunities for technical refinement. The reported gains are in light sensitivity and in simple object tasks, not in a general return of sight. Those are the limits of what the study shows.

For people with advanced retinitis pigmentosa, the gain today is a treatment that has passed a safety test in patients, along with a foundation for further work. It is not yet a product. The researchers give no date for a larger trial or for regulatory review, and the reports do not state a price.
